CLINICAL SIGNIFICANCE:

Anti-Streptolysin O, an antigen produced by Lancefield Group A Streptococci. ASO Titres are elevated in the sera 80% to 85% of patients with rheumatic fever and in 95% of patients with acute Glomerulonephritis. The World Health organization recommends the use of ASO to aid the diagnosis of Streptococcal Infections. Increased Levels: are observed in scarlet fever, Acute Rheumatic Arthritis, Tonsillitis & other various streptococcal infections.

PRINCIPLE:

The reagent consists of suspension of latex particles homogenous size sensitized with anti- ASO, capable of aggregation in the presence of ASO. This aggregation process produces an increase in the size of latex particles which in turn produces an increases in absorbance of the system.

SHELF LIFER:

eagents are stable up to 2-8°C intill the expiration datestated on the bottle for unopened Bottles. Protect the reagent from light and contamination.

SAMPLE:

Fresh sera or stored at 2-8°C for no longer than 48hrsIt is necessary to freeze the sample when the assay isto be carried out after that period of time. Discard contaminated and hemolyzed sample.

WORKING REAGENT PREPATION:

Prepare working reagent by mixing 4 parts R1-ASO & 1 Part of R2- ASO. Stability of working reagent is stable for 7 days at 2-8°C.

REAGENT:

Ready to use, Two Reagents system.

CALIBRATION:

Calibration type : Spline (5 points) Prepare the dilutions of calibrators in given belowmanner for calibration.

PROCEDURE :

The samples and reagents should be brought to room temperature prior to use.

LIMITATIONS & PRECAUTIONS:

● Storage conditions as mentioned on the kit tobe adhered. ● Do not freeze or expose the reagents to highertemperature as it may affect the performance of thekit. ● Before the assay bring all the reagents to theroom temperature. ● Avoid contamination of the reagent during assayprocess. ● Use clean glassware free from dust and debris.● Reagent to sample ratio as mentioned abovemust be strictly adhered, as any changes itaffects the factor and result.

PERFORMANCE CHARACTERISTIC:

  • SENSITIVITY: 10 IU/mL
  • LINEARITY:Reagent is linear up to 800 IU/ml. For high value samples, dilute samples with 0.9% saline Repeat the test & multiply the results with dilution factor in end.
  • INTERFERENCES:No significant interactions were observed by Bilirubin,(171umol/L), Hemoglobin(5g/L), Triglycerides (2.28 mmol/L), RF (210IU/ml).Other drugs may interfere in the tests.

QUALITY CONTROL:

To ensure adequate quality control, it is recommended that the laboratory should use a normal and abnormal commercial reference control serum. Please note that the quality control material is used to check the function of reagents and the machine together.

REFERENCE NORMAL VALUE:

Serum Adults: Up to 200 IU/ml.
